Photoreceptor disc morphogenesis: The classical evagination model prevails
نویسنده
چکیده
Vision begins in photoreceptor outer segments with light captured by opsins in continually synthesized disc membranes. The process by which rod photoreceptor discs are formed has been controversial. In this issue, Ding et al. (2015. J. Cell Biol. http://dx.doi.org/10.1083/jcb.201508093) show conclusively that rod discs are formed by plasma membrane evagination.

Peripherin diverts ciliary ectosome release to photoreceptor disc morphogenesis
Formation of membrane discs in photoreceptor cells requires evagination of its ciliary plasma membrane by an unknown molecular mechanism. Salinas et al. (2017. J. Cell Biol. https://doi.org/10.1083/jcb.201608081) show that peripherin (also known as peripherin-2 or peripherin-2/rds) diverts membrane traffic to photoreceptor disc formation by inhibiting ectosome release from the cilium.

Electron microscopic examination of the bases of adult rod and cone outer segments (rhesus monkey, ground squirrel, and grey squirrel) has led to a new model of disc morphogenesis. In this model the disc surfaces and disc rims develop by separate mechanisms and from separate regions of the membrane of the inner face of the cilium. This membrane is alternately specified into regions that will fo...
متن کاملCytochalasin D disrupts outer segment disc morphogenesis in situ in rabbit retina.
Exposure of rabbit retina to cytochalasin D (CD) via a single intraocular injection results in basal rod outer segment (ROS) and cone OS (COS) discs with abnormally large diameters; the overgrown OS membranes extend along either the cell outer or inner segment. Twenty-four hours after the injection, basal ROS and COS discs appear to have recovered their normal diameter, indicating the reversibi...

متن کاملDiscs of mammalian rod photoreceptors form through the membrane evagination mechanism
Photoreceptor discs are membrane organelles harboring components of the visual signal transduction pathway. The mechanism by which discs form remains enigmatic and is the subject of a major controversy. Classical studies suggest that discs are formed as serial plasma membrane evaginations, whereas a recent alternative postulates that discs, at least in mammalian rods, are formed through intrace...
